The Enzymic Synthesis of Amino Acyl Derivatives of Ribonucleic Acid I. THE MECHANISM OF LEUCYL-, VALYL-, ISOLEUCYL-, AND METHIONYL RIBONUCLEIC ACID FORMATION”

چکیده

The enzymic formation of enzyme-bound amino acyl adenylates from adenosine triphosphate and amino acid (Equation 1) has been recognized for several years (l-8) and enzymes specific for certain of the amino acids have been isolated in a number of laboratories (2, 9, 10). These same enzymes are now known (11-15) to catalyze a second reaction involving the transfer of the amino acyl moiety from the adenosine phosphate moiety to a specific type of ribonucleic acid (Equation 2). The over-all reaction catalyzed by such amino acyl ribonucleic acid synthetasesl is summarized in Equation 3.

Changes in transfer ribonucleic acids accompanying encystment in Acanthamoeba castellanii.

Transfer ribonucleic acid (tRNA) from exponentially growing cells (trophozoites) and from precysts of Acanthamoeba castellanii were examined by reversed-phase column (RPC-2) chromatography. This system gave excellent resolution of isoaccepting species of tRNA. The tRNAs for 12 amino acids were studied. A comparison of trophozoite and precyst tRNA elution profiles revealed no apparent difference...

Proofreading in vivo: editing of homocysteine by methionyl-tRNA synthetase in Escherichia coli.

Previous in vitro studies have established a pre-transfer proofreading mechanism for editing of homocysteine by bacterial methionyl-, isoleucyl-, and valyl-tRNA synthetases. The unusual feature of the editing is the formation of a distinct compound, homocysteine thiolactone. Now, two-dimensional TLC analysis of 35S-labeled amino acids extracted from cultures of the bacterium Escherichia coli re...

Homology of yeast mitochondrial leucyl-tRNA synthetase and isoleucyl- and methionyl-tRNA synthetases of Escherichia coli.

Respiratory deficient mutants of Saccharomyces cerevisiae previously assigned to complementation group G59 are pleiotropically deficient in respiratory chain components and in mitochondrial ATPase. This phenotype has been shown to be a consequence of mutations in a nuclear gene coding for mitochondrial leucyl-tRNA synthetase. The structural gene (MSL1) coding for the mitochondrial enzyme has be...

Stoichiometry and composition of an aminoacyl-tRNA synthetase complex from rat liver.

The particulate aminoacyl-tRNA synthetases of rat liver were copurified about 1000-fold with more than 20% yields for individual synthetase activities. Measurements of aminoacylation activities showed that lysyl-, arginyl-, leucyl-, isoleucyl-, and methionyl-tRNA synthetases in the purified complex cosedimented at 18 S. The molecular weight of the synthetase complex is about one million, as est...

Alternative pathways for editing non-cognate amino acids by aminoacyl- tRNA synthetases

Evidence is presented that the editing mechanisms of aminoacyl-tRNA synthetase operate by two alternative pathways: pre-transfer, by hydrolysis of the non-cognate aminoacyl adenylate; post-transfer, by hydrolysis of the mischarged tRNA. The methionyl-tRNA synthetases from Escherichia coli and Bacillus stearothermophilus and isoleucyl-tRNA synthetase from E. coli, for example, are shown to rejec...
